De Nyse was born in Brooklyn approximately in 1841 to Denyse H. De Nyse and his wife Maria. He was from the "well-known New Utrecht family."〔"Mrs. Sarah De Nyse" (obituary), ''Brooklyn Daily Eagle'', November 11, 1899, p. 16.〕 He attended public school in New York City.〔''Fourteen Annual Report of the Board of Education of the City and County of New York for the Year Ending January 1, 1856'' (New York: Wm. C. Bryant & Co., 1856). De Nyse is listed in "Schedule III", p. 40 and 66.〕
